چکیده

Our previous studies have demonstrated that surfaceactive copolymers of Pluronic® surfactants and tertiary amino polycations such as poly (2-dimethylaminoethyl methacrylate) (PDMA) possess an enhanced DNA transfection efficiency compared to PDMA homopolymers. In order to understand the mechanism of such action, we conducted model studies of the interaction of polymer with phospholipid vesicles and cell membranes. Three cationic methacrylate polymer systems were used in this study: PDMA homopolymer, and PEO-b-PDMA and Pluronic L92-b-PDMA block copolymers. The results showed that PDMA and PEO-b-PDMA only interact with the surface of the vesicles, whereas L92-b-PDMA interacts strongly with the inner hydrophobic segment of the vesicles. The studies show that the strong interaction between L92-b-PDMA and phospholipid membrane could be used to explain the enhanced transfection efficiency previously shown by the Pluronic L92-b-PDMA copolymer.

Pentablock copolymers of pluronic F127 and modified poly(2-dimethyl amino)ethyl methacrylate for internalization mechanism and gene transfection studies

Cationic polymers are one of the major nonviral gene delivery vectors investigated in the past decade. In this study, we synthesized several cationic copolymers using atom transfer radical polymerization (ATRP) for gene delivery vectors: pluronic F127-poly(dimethylaminoethyl methacrylate) (PF127-pDMAEMA), pluronic F127-poly (dimethylaminoethyl methacrylate-tert-butyl acrylate) (PF127-p(DMAEMA-t...

Synthesis and Properties of Novel Cationic, Temperature-Sensitive Block-Copolymers

Facile, one-step synthesis of self-assembling, cationic block copolymers of poly(2-N-(dimethylaminoethyl) methacrylate) (pDMAEMA) and PEO-PPO-PEO (Pluronic®) is developed. The copolymers are obtained via free-radical polymerization of DMAEMA initiated by Pluronic-radicals generated by cerium (IV). The copolymers possess surface activity, are polycationic at pH<7.1, and self-assemble into micell...
